How to use the D/E Neutralizing Broth

Here are general instructions for using the DE Broth Dry Speci-Sponge Bag with sterile gloves in a food laboratory setting:

Materials Needed:

Step-by-Step Instructions:

  1. Prepare the Work Area:
    • Clean and disinfect the surface where you will be handling the samples.
    • Gather all necessary materials and ensure the area is organized to avoid contamination.
  2. Put on Sterile Gloves:
    • Carefully put on the sterile gloves provided in the kit.
    • Avoid touching any non-sterile surfaces with the gloves once they are on.

  3. Hydrate the Sponge:
    • Open the Speci-Sponge Bag by tearing or cutting it at the top, being careful not to touch the inside of the bag.
    • Add the appropriate amount of sterile diluent (usually specified by your laboratory protocol) to the sponge. This is typically done to fully hydrate the sponge and activate the DE broth.
    • Allow the sponge to soak for 1-2 minutes to ensure it is fully saturated.

  4. Sample Collection:
    • Once the sponge is hydrated, carefully remove it from the bag without contaminating the outer surface.
    • Use the sponge to swab the designated area, applying firm and consistent pressure as you move it across the surface. Ensure you cover the area thoroughly to collect a representative sample.

  5. Return the Sponge to the Bag:
    • After collecting the sample, carefully place the sponge back into the Speci-Sponge Bag.
    • Seal the bag securely to prevent contamination or leakage.

  6. Label the Sample:
    • Clearly label the bag with relevant information such as sample ID, date, time, and any other details required by your laboratory's protocol.

  7. Transport and Storage:
    • If not processed immediately, store the sample in a cool environment, typically refrigerated at 2-8°C.
    • Transport the sample to the testing area or laboratory as soon as possible to maintain sample integrity.

  8. Dispose of Used Materials:
    • Dispose of gloves and any other disposable materials according to your laboratory's biohazard waste protocols.
    • Clean and disinfect your work area after completing the sample collection process.
